چکیده

Purpose:Most studies characterizing antitumor properties of invariant natural killer T (iNKT) cells have used the agonist, a-galactosylceramide (a-GalCer). However, a-GalCer induces strong, long-lasting anergy of iNKT cells, which could be a major detriment for clinical therapy. A novel iNKT cell agonist, b-mannosylceramide (b-ManCer), induces strong antitumor immunity through a mechanism distinct from that of a-GalCer. The objective of this study was to determine whether b-ManCer induces anergy of

β-mannosylceramide activates type I natural killer t cells to induce tumor immunity without inducing long-term functional anergy.

Mouse and human iNKT cell agonist β-mannosylceramide reveals a distinct mechanism of tumor immunity.

Type 1 or invariant NKT (iNKT) cell agonists, epitomized by α-galactosylceramide, protect against cancer largely by IFN-γ-dependent mechanisms. Here we describe what we believe to be a novel IFN-γ-independent mechanism induced by β-mannosylceramide, which also defines a potentially new class of iNKT cell agonist, with an unusual β-linked sugar. Like α-galactosylceramide, β-mannosylceramide dire...

The Efficacy of Toll-Like Receptors in Awakening Dendritic Cell/Natural Killer Cell System for Eradication of Tumors

Natural killer (NK) cells are effector cells of the innate immune system that exert direct cytotoxic functions. Ubiquitously-expressed toll-like receptors (TLRs) have been recognized as one of the major components promoting dendritic cell (DC) maturation, which may induce polarized immune responses beneficial to cancer immunotherapy. TLR-activated NK cells and DCs are prerequisite for robust ac...

Synergistic induction of adaptive antitumor immunity by codelivery of antigen with α-galactosylceramide on exosomes.

Exosomes and the invariant NKT (iNKT) immune cell ligand α-galactosylceramide (αGC) may offer novel tools for cancer immunotherapy. In this study, we investigated whether exosomes loaded with αGC can activate iNKT cells and potentiate a cancer-specific adaptive immune response. αGC loaded exosomes readily activated iNKT cells both in vitro and in vivo. Exosomes loaded with αGC plus the model an...

Memory type 2 helper T cells induce long-lasting antitumor immunity by activating natural killer cells.

Functionally polarized helper T cells (Th cells) play crucial roles in the induction of tumor immunity. There is considerable knowledge about the contributions of IFN-producing Th1 cells that supports the role of cytotoxic cluster of differentiation (CD8) T cells and natural killer (NK) cells, but much less is known about how IL-4-producing Th2 cells contribute to tumor immunity. In this study,...
